That is something I really would like to do, coupled with the Pan America Highway:-
I have already done part of it having driven (15 years ago) over some of Ecuador Highway 35 (E-35), "Troncal de la Sierra" (Highland's Road). The surface is quite good in some parts but in the province of Cañar it's pretty iffy. What is amazing is that you are driving along a good road, which suddenly stops, and you detour from 4 lane tarmac to a jungle track. When I was there there were toll booths, but they had been abandoned as there wasn't the volume of traffic to justify staffing them, or completing the road.
If I bought a new D4 in Ushuaia and drove it to Anchorage (Alaska) what price could I sell it for?
A pipe dream I fear.

There is a video about it here from Jay Leno's programme, I'm pretty sure Land Rover also posted a video about the trip but can't find that at the moment
It's a journey I would love to do Cheers Ian
