Yes, you'll need to disconnect the door stay from the body as they are one whole piece and then reconnect once you've put the seal past it before sticking in place otherwise the door will shut on you. The seal goes a particular way (it has a 'g' shape profile) so make sure you get this right otherwise you will have to take it off again.
Worth using masking tape to remind you of the shape of the old seal particularly on the sides so that it goes back in the same place as it doesn't follow the edges of the door.
Start at the bottom and it should take about 10 minutes

It depends. Are you talking the one on the door or the body? The body is easy to fit without removing anything. I replaced the drives one on mine recently as it was worn from rubbing on entry and exit. If it's the one on the door itself, then as above.

Duckworth parts?
Matlock Land Rover?
I would only fit a genuine seal
Watch the metal trim clips on the bottom plastic section when you lift it, sometimes they can ping off into the cavity below and are a pig to fish out

Just done my drivers side one, 10 minute job, as above make sure the corners go in the right place and you fit it under the kick plate properly but once done you’ll notice a reduction in wind noise! I want to see the sweets before I get into your windowless van... I'm not stupid!
